Die 2. U-Flottille wurde am 01.09.1936 unter dem Namen U-Flottille SALTZWEDEL als Ausbildungs-, später Frontflottille, in Wilhelmshaven aufgestellt. Nach dem 31.12.1939 wurden die bestehenden Flottillen zusammengelegt. Die 2. U-Flottille SALTZWEDEL, und die 6. U-Flottille HUNDIUS wurden zur 2. U-Flottille vereinigt. Die U-Flottille SALTZWEDEL bekam die Feldpostnummer M-25 522.
